Isolation of catenated and replicating DNA molecules of colicin factor E1 from minicells.

چکیده
Various catenated and replicating molecules of colicin E1 isolated from minicells have been identified in regions of neutral sucrose density gradients or cesium chloride-ethidium bromide density gradients. These colicin molecules have been found in those regions of the gradient where pulse-labeled DNA accumulates.

Replication of colicin E1 plasmid DNA in cell extracts. Origin and direction of replication.
The structures of molecules of colicin E1 plasmid DNA that were in the process of being replicated in vitro were examined electron microscopically. Circular molecules containing a loop of approximately 7% of the length of the molecules were the major class of replicating molecules. The location of the loop was determined by treating the molecules with restriction endonuclease EcoR1, which intro...
متن کاملConstruction and characterization of new cloning vehicles. III. Derivatives of plasmid pBR322 carrying unique Eco RI sites for selection of Eco RI generated recombinant DNA molecules.
In vitro recombinant DNA techniques were used to construct two new cloning vehicles, pBR324 and pBR235. These vectors, derived from plasmid pBR322, are relaxed replicating elements. Plasmid pBR324 carries the genes from pBR322 coding for resistance to the antibiotics ampicillin (Apr) and tetracycline (Tcr) and the colicin E1 structural and immunity genes derived from plasmid pMBI. Plasmid pBR32...
متن کاملIsolation and characterization of mutants of colicin plasmids E1 and E2 after Mu bacteriophage infection.
Cells colicinogenic for the colicin plasmids E1 or E2 (Col E1 and Col E2, respectively) were selected for a loss of colicin production after infection with bacteriophage Mu. Extrachromosomal deoxyribonucleic acid that was larger than the original colicin plasmids was found in such cells. A small insertion mutant in Col E1 deoxyribonucleic acid affecting active colicin production without affecti...
متن کاملSegregation into and replication of plasmid deoxyribonucleic acid in chromosomeless segregants of Escherichia coli.
The progeny cells of Escherichia coli strain P678-54, which normally do not contain deoxyribonucleic acid (DNA), were found to carry DNA when the parental cell carried colicin factor E1 (Col E1). The DNA found in the normally DNA-less segregants was shown to be Col E1 DNA, which is present primarily as a covalently closed circular molecule that can undergo more than one complete cycle of replic...
متن کاملOrigin of replication of colicin E1 plasmid DNA.
Cleavage maps of colicin E1 plasmid DNA and its smaller derivative, pNT1 DNA, were constructed by using restriction endonucleases. The nucleotide sequence of a region that contains the orgin of replication was determined. The site of the nucleotide from which DNA replication is initiated was determined with 6S L-fragments, the DNA fragment first made on colicin E1 plasmid DNA. The fragments wer...
